DescriptionThe E. coli O145 antigen monomer consists of a linear trimer with the structure: [->4)aNeu5Ac(2->3)aLFucNAm(1->3)bDGlcNAc(1->], where Neu5Ac is N-acetyl-neuraminic acid and L-FucNAm is 2-acetamidoylamino-2,6-dideoxy-L-galactose. The LFucNAm transferase (WbuB) is 98% identical to enzymes in the O151 and O118 antigens where the same linkage is O145 is found. These are, in turn members of a clade of such enzymes carrying out the same linkage (with either LFucNAc or LFucNAm), always at the end of the monomer chain, in antigens O4, O26, O29, O118, O145, O151, and O172. The WbuW enzyme is very divergent from other known glycosyltransferases, consistent with the rare aNeu5Ac(2->3)LFucNAm reaction that it must be carrying out as inferred by the process of elimination.
